Marvel's Spider-Man 2 is set 10 months after the events of Spider-Man: Miles Morales (2020). As with previous installments in the series, the game features a large ensemble cast of various characters derived from the history of Spider-Man comics and other media. Yuri Lowenthal and Nadji Jeter reprise their respective roles as protagonists Peter Parker / Spider-Man and Miles Morales / Spider-Man from Spider-Man (2018) and Spider-Man: Miles Morales. Other cast members reprising their roles include Laura Bailey as Mary Jane Watson, Darin De Paul as J. Jonah Jameson, Jacqueline Piñol as Rio Morales, Griffin Puatu as Ganke Lee, Mark Rolston as Norman Osborn, and Stephen Oyoung as Martin Li / Mister Negative. Tony Todd provides the voice and motion capture for Venom, an extraterrestrial symbiote capable of bonding to human bodies as hosts. Jim Pirri provides the voice and motion capture for the character Sergei Kravinoff / Kraven the Hunter, the leader of a faction of mercenaries called the "Hunters". Their members include Maxwell Markham / Grizzly, and they seek to hunt superpowered beings, including the Spider-Men. Tara Platt will reprise her role from Spider-Man as Yuri Watanabe, a disgraced former police captain and ally of Peter Parker, who now assumes the vigilante identity of "Wraith" following the events of the game's DLC campaign, The City That Never Sleeps. Mark Whitten portrays Curt Connors / Lizard, a former Oscorp scientist who turned himself into a giant reptilian creature;Connors previously appeared in the mid-credits scene of Miles Morales. Additional antagonists who were previously referenced in prior entries make full appearances throughout the game's story, including Flint Marko / Sandman, a lowly burglar who through an accident, gained the ability to turn into molecules of sand and manipulate them at will, and Quentin Beck / Mysterio, a visual effects artist leveraging virtual reality to create deadly illusions. Harry Osborn, now motion captured and voiced by Graham Phillips after originally being voiced by Scott Porter in Spider-Man, will appear in the game in a more prominent role after making brief cameo appearances in both Spider-Man and Miles Morales.
